摘 要:本实验以干槐花为原料,研究PEG4000的质量分数为20%、22%、24%、25%、30%、35%、40%,(NH4)2SO4的质量分数为20%、25%、26%、28%、30%、35%、40%、43.5%、45%,pH为3.13、4、5、6、7.35、8.36和温度为30℃、40℃、50℃、60℃、70℃、80℃、90℃、100℃槐花中蛋白质的双水相萃取及氨基酸组成。以PEG4000质量分数、(NH4)2SO4质量分数、pH、温度为单因素做四因素三水平的正交实验来确定最佳萃取条件。结果表明PEG4000的质量分数为30%、(NH4)2SO4的质量分数为45%、pH为5、温度为40℃为最佳萃取条件,且在最佳条件下的下相萃取率达到92.14%。最佳条件下的氨基酸组成为异亮氨酸、亮氨酸、赖氨酸、胱氨酸、酪氨酸、精氨酸、天冬氨酸、甘氨酸、丝氨酸、谷氨酸、苏氨酸、缬氨酸、甲硫氨酸、苯丙氨酸、组氨酸、丙氨酸、脯氨酸。

Abstract:PEG4000 are studied in this experiment use dry sophora japonica as a raw material, the mass fraction of 20%, 22%, 24%, 25%, 30%, 35%, 40%, mass fraction of (NH4)2SO4 was 20%, 25%, 26%, 28%, 30%, 35%, 40%, 43.5%, 45%, pH 3.13, 4, 5, 6, 7.35, 8.36 and the temperature of 30 ℃, 40 ℃, 50 ℃, 60 ℃, 70 ℃, 80 ℃, 90 ℃, 100 ℃ sophora japonica in the aqueous two-phase extraction and the amino acid composition of protein. With PEG4000 mass fraction and mass fraction (NH4)2SO4, pH and temperature for single factor four factors three levels orthogonal experiment to determine the optimal extraction conditions. Results show that the mass fraction of 30% PEG4000 and mass fraction of (NH4)2SO4 45%, pH 5, temperature of 40 ℃ for the optimum extraction conditions, and under the best conditions of extraction rate reached 92.14%. Best under the condition of amino acid composition of isoleucine, leucine, lysine, cystine, tyrosine, arginine and aspartic acid, glutamic acid, glycine, serine, threonine, valine, methionine, phenylalanine, histidine, proline and alanine.
