摘要:以生姜粉为原料,通过高效液相色谱(HPLC)测定分配系数法优选高速逆流色谱(HSCCC)分离的最佳溶剂体系,确定了最佳的HSCCC分离条件:溶剂体系为正己烷-乙酸乙酯-甲醇-水(5.5:5:5.5:5,v/v)以及同种体系的(6.5:5:6.5:5,v/v),流速2 mL/min、转速800 r/min、分离温度25℃,经梯度洗脱即可从生姜提取物中分离得到4种单体组分,经HPLC分析为6-姜酚,6-姜烯酚,8-姜酚三种单体成分,纯度较高,还得到只含有8-姜酚和10-姜酚的混合姜酚。

   对分离得到的6-姜酚,6-姜烯酚,8-姜酚三种单体成分进行了抗氧化性分析,结果表明具有一定的清除羟基自由基和DPPH自由基能力,清除能力与浓度呈较明显的量效关系;稳定性实验表明,姜酚在非碱性条件下对温度、时间和光照等具有较好的稳定性,适用于食品生产。

Abstract:Ginger powder as raw material, by high-performance liquid chromatography (HPLC) determination of the partition coefficient method is preferably high-speed countercurrent chromatography (HSCCC) the separate solvent system to determine the best HSCCC separation conditions: solvent system of n-hexane - ethyl acetate- methanol - water (5.5:5:5.5:5, v/v) and the same kind of system (6.5:5:6.5:5, v/v), flow rate of 2 mL/min, the rotational speed of 800 r/min, separation temperature25 ℃, gradient elution can be separated from the ginger extract monomer component analyzed by HPLC for 6-gingerol, 6-Shogaol 8-ginger phenols three monomer composition, purity than thealso contains only 8-gingerol and 10-gingerol mixed gingerol.

Isolated 6-gingerol, 6-Shogaol, 8-gingerol three monomer components of the antioxidant activity analysis, the results showed that the scavenging DPPH radical scavenging capacity and concentration showed an obvious dose-effect relationship; stability experiments showed that gingerol has good stability under alkaline conditions of temperature, time and light, suitable for food production.
