摘要:以芦荟为原料,通过单因素和正交试验查找到芦荟多糖提取液脱蛋白、脱色等工艺的最佳条件。用蒽酮-硫酸法绘制标准曲线。通过四种常见的脱蛋白方法进行比较,结果证明明木瓜蛋白酶-Sevag法除蛋白效果最好,蛋白清除率高达91%。通过正交实验确定了最佳提纯工艺条件。结果表明:DA201-C大孔树脂对芦荟多糖的吸附能力最强,最佳树脂用量是25 g,最佳速度是0.5 mL/min,最佳溶液pH为 5.5。对芦荟多糖溶液进行了羟基自由基和DPPH自由基清除实验研究。实验结果表明,在一定范围内,当多糖浓度较高时,羟基自由基和DPPH自由基的清除率就好越高。实验表明芦荟多糖是很好的天然抗氧化剂。

Abstract: With aloe as material, we can try many factors and orthogonal test to make sure the best condition of technology of deproteinization, macroporous resin adsorption. we can draw standard curve with the methods of Anthrone- sulfuric acid . The results show that papain - Sevag method is the best to remove protein by comparing four common protein method ,and protein removal rates as high as 91%. Through orthogonal experiment to find the best purification process conditions though investigating type of macroporous resin, resin, slurry concentration, the adsorption velocity .The results show that: D201-C Amacroporous resin has the better adsorption abilities. When the weight of D201-C was 25g,the moving speed was 0.5mL/min, the pH was 5.5 ,the result of purification is the best. scavenging hydroxyl radical and DPPH radical of aloe polysaccharide,the result show that when the concentration of polysaccharide from aloe is higher with limits the clearance rate is higher. Experiments show that aloe polysaccharide is a good natural antioxidant.
