摘要:窑湾甜油出现于清康熙年间。中医认为,甜油入肾经,可滋阴养胃。窑湾甜油中的霉菌能产生大量淀粉酶,其对甜油中的发酵盛传具有重要作用。
淀粉酶指的是一类能够将催化淀粉水解和转化的酶的总称。同时,淀粉酶也是一种用途极为广泛的生物催化剂,能催化淀粉向葡萄糖和麦芽糖的转变,可应用于淀粉的糖化和液化、造纸业、清洁剂工业、纺织品的脱浆、化学工业、制药工业和临床医学的分析等等。
本研究从徐州窑湾甜油中分离筛选出9株霉菌,并通过一系列初筛和复筛的方法得到一株淀粉酶活性最高的菌株命名为M8。进一步通过对其产酶条件的优化和酶学性质的研究,确定了M8生产淀粉酶的最优发酵条件和淀粉酶的性质。最终初步确定霉菌所产淀粉酶的酶活的最适菌株培养温度为28℃,酶活测定的最适处理温度为80℃,培养液的最适pH为6.5,酶缓冲液最适pH为6.6,以及最适接种量为1.0%,并确定了Ca2+对该菌株所产的淀粉酶是有一定促进作用而某些有机溶剂对其发挥催化作用是有不利影响的。本研究结果对窑湾甜油发酵工艺的改进,提高产品的竞争力具有重要意义。

Abstract:Yao Wan Sweet Oil appears in the Kangxi period of the Qing Dynasty.The mould from Yao Wan Sweet Oil can produce amylase which plays an important role in the Yao Wan Sweet Oil.
Amylase is a general name of a kind of enzyme which can catalyze the hydrolysis of starch and transformation. Amylase is a widely used biocatalyst which can catalyze the starch to glucose and maltose.
In this study, soil samples were obtained from XuZhou Yao Wan Sweet Oil and saved in sterile sample bags.9 strains of mould was screened. Four strains with pectinase activity were obtained and with the highest pectinase activity was named M8.The amylase was assayed by DNS method. Then the enzymatic properties of M8 were studied. And the most appropriate training temperature is28℃, the most handling temperature is 80℃,the most culture pH is 6.5, the most buffer pH is 6.6, and the most amount of inoculum is 1%. And it was determined Ca2+ play an important role in promoting of the amylase which is produced by the strain. At the same time, some organic solvents play a catalytic role in enzyme catalysis. This research has profound meaning for the improvement of Yao Wan Sweet Oil and the products competitive.
